Concentration of Megaloblasts: An Aid in the Diagnosis of Pernicious Anaemia.

چکیده

The buffy coat in a haematocrit tube consists of white cells, which, in spite of their larger size, fall with less velocity than the smaller red cells. This consideration suggested that the bigger, nucleated red cells might be found in increased numbers in or immediately beneath the white cell layer, and this has proved to be the case. Incidentally, both macrocytes and microcytes are found concentrated in this agea, thus exaggerating the typical pernicious anaemia picture. A further point of interest is the study of the white cells themselves, which in ordinary films are very scarce, owing to the generally occurring leucopenia. Cooke's Type I and II cells can thus be conveniently studied.

Pernicious anaemia.

DEFICIENCY of vitamin B12 (cyanocobalamin) in man results in a macrocytic animia, leucopa2nia, the appearance of megaloblasts and giant band cells in the marrow, reduced formation of platelets, and also-changes in the peripheral and central nervous systems. In temperate climates the most common cause of vitamin B12 deficiency is pernicious anaemia. This is by no means a rare disease, having an ...

Sideroblastic and megaloblastic change.

Introduction THE simultaneous occurrence in the bone marrow of megaloblasts and ring-sideroblasts may result from a variety of causes. In a series of seventy patients with sideroblastic anaemia, MacGibbon and Mollin (1965) observed megaloblastic change in 544O of cases of primary sideroblastic anaemia, and in 74YO of cases of secondary sideroblastic anaemia. The megaloblastic change was attribu...

Minimal criteria for the diagnosis of megaloblastic anaemia of pregnancy.

The routine examination of buffy coat films is a valuable diagnostic aid to screening large numbers of specimens of blood and is applicable to hospital and general practice, provided that a bottle of sequestrinated venous blood can be examined in the laboratory within 24 hours. The diagnosis of folic-acid deficiency anaemia of pregnancy can be made, in many cases, before the appearance of megalo...

Megaloblasts in the Peripheral Blood

THE finding of megaloblasts in the peripheral blood is often regarded as an uncommon event, confined to patients with severe anaemia. Methods of concenitrating the megaloblasts by making preparations from the "buffy coat'i of the haematocrit have been described (Goodall, 1957, Whitby and Britton, 1957). This paper records our experience of examining the peripheral blood for megaloblasts. METHOD...
